The Applied Biosystems PrioCHECK African Swine Fever Virus Ab Kit is an easy‐to‐use ELISA that enables detection of antibodies directed against the p30 protein of African swine fever virus (ASFV) in porcine serum. The p30 protein was selected as the indicator of virus exposure in this kit due to its strong antigenicity, its expression at early stage of infection, and its high conservation among ASFV genotypes.

African swine fever, caused by ASFV, is a highly contagious, lethal disease in pigs and wild boars. ASFV spreads rapidly in pig populations through direct or indirect contact and the bite of infected soft tick species. Outbreaks of ASFV cause severe economic losses in pig-producing countries, making rapid detection of infected pigs essential to control the disease. Given the long‐term persistence of anti‐ASFV antibodies after infection, serology tests provide an accurate and easy approach to control the health status of porcine herds and wildlife (wild boars).

Test procedure
p30 protein is immobilized on the test plate. Sera are added to the plate where any p30 antibodies in the sera bind to the p30 protein on the plate. After a washing step, an anti-swine immunoglobulin coupled to HRP is added. After a second washing step, TMB chromagen substrate is dispensed into the wells to react with any HRP present and produce color. The presence of color is measured optically at a wavelength of 450 nm and indicates the presence of antibodies directed against the p30 protein of ASFV in the serum sample.
